In contrast to traditional dyeing, there is no reaction between the sample and the dye solution, so the sample is not colored. A white sample with a low electron density can be contrasted with a black background, forming an obvious negative contrast in contrast to the density difference between the sample and the dye. Using specialized software such as RELION, the particles were pretreated and selected, and the conformation of the target complex with low resolution was obtained through rounds of 2D average classification and optimization.
